One thing I am sure of is everything will be better.

而不能说：

One thing of which I am sure is everything will be better.

不知你是从哪里看到的。你这个句式两个说法都可以。当然英语中有些是不行的。如：You are the student I'm looking for. 这里你不能说for whom I'm looking. 严格讲，你如果用了of which，说明是比较正式的文体，表语从句最好有that. 不过，这不算什么错误。
